Influence of Intracellular Reactive Oxygen Species in Several Spermatozoa Activity in Indonesian Ongole Bull Cryopreserved Sperm

Certain factors may affect sperm motility and integrity in a freezing process. Reactive Oxygen Species (ROS) is essential damaging the sperm, as previous study revealed that ROS major cause membrane of spermatozoa. This aimed to understand production its interaction within motility, mitochondrial activity DNA damage cryopreservation process Indonesian Ongole grade bull. A total 40 ejaculated semen samples from four different bulls had taken. The intracellularly detected by 2 -7 -Dichlorodihydrofluorescein Diacetate (DCFH-DA) propodium iodide (PI). Mito Tracker Red kit was being used discover activity, while sperms' Terminal Deoxynucleotidyl Transferase dUTP Nick End Labeling (TUNEL). Examinations were done under fluorescence microscope. Results demonstrated significant increase fresh semen, diluted or equilibrated before frozen (at 12°C 4°C). showed no differences between frozen-thawed thawed decrease shown due equilibration which temperature significantly higher at 4°C than temperatures, however, diluting not given any causes damage. concluded with effects might be main factor leads decreasing but effect discovered on occurred. results have proven stated has mechanisms affecting integrity. It also comes new theory affected chemical diluter despite thawing
